چکیده

A series of experiments was designed determining the seeding and ultimate to study the effect of spleen size on retention of CFU in the spleen of a the seeding and retention of spleen recipient mouse is the size of the colony-forming units (CFU) after inspleen. It is concluded that the most jection into irradiated mice. The cellureasonable value of the fraction’ of larity of the spleens of recipient mice injected CFU that produces spleen was varied using a variety of expericolonies should be determined from mental procedures, and the resulting the number of CFU recoverable from spleen colony production was anathe fully shrunken spleen. In these lyzed with respect to CFU seeding. mice, the fraction was 9.5#{176}/o. It was found that the major factor T HE FRACTION (f) of spleen colony-forming units (CFU) injected into a lethally irradiated mouse that produces hemopoietic colonies in the spleen has usually been determined by the secondary transplantation method of Siminovitch et al.’ Several more recent reports have shown that the number of CFU that can be recovered from the spleen of an irradiated animal following an injection of hemopoietic cells depends on the interval of time between the injection and the assay.2’4 One of these3 demonstrated that the number of CFU recoverable is directly related to the number of cells in the spleen at the time of assay. The explanation suggested for this relationship was that after injection CFU enter the spleen, but as the spleen collapses due to the radiation, the CFU are expelled or destroyed. It was concluded that the calculation of the f number should be based on the number of CFU recoverable from the fully shrunken spleen, rather than on the maximum numbers recoverable, i.e., about 2 hr after injection. It had earlier been suggested5 that the initial decrease (after about 2 hr) in the number of CFU per spleen was due to an inability to extract and disperse all the CFU from the spleen. This same suggestion has been enforced more recently by Till and McCulloch,6 who have reported experiments in which colony production in W/W” mice receiving different doses of radiation (including no radiation) remained the same. In an attempt to clarify the fate of CFU entering the spleen after intravenous injection, a variety of experiments have been carried out and are reported in this paper.

Transplantation and Homing of Mouse Embryonic Stem Cells Treated with Erythropoietin in Spleen and Liver of irradiated mice

Background: The present study was designed to evaluate the homing potential of mouse embryonic stem cells (ESC) treated with erythropoietin (EPO) in hematopoietic organs such as spleen and liver after transplantation using morphological and immuno-histochemical techniques. Methods: Day-four embryoid body (EB)-derived cells were dissociated and re-plated in medium in the presence and absence of ...

THE INTERACTION OF RADIATION AND INTERCALATING AGENTS IN NORMAL BONE MARROW CELLS AS EVALUATED BY SPLEEN COLONY ASSAY TECHNIQUE: THE EFFECTS OF BLEOMYCIN SULFATE AND ACT INOMYCIND.

The relationship between the way in which normal hemopoietic stem cells respond to irradiation alone or in the presence of bleomycin sulfate (BLM-S) and actinomycin 0 (ACT-D) was investigated. Single doses of BLM-S at 0.3 mg/kg and ACT-O at 0.10 mg/kg body weight were injected intravenously 1-6 hours prior to whole body irradiation and treatment was repeated twice more with time intervals....

Nude mice bearing human CSF-producing tumor: analysis of hemopoietic factor(s) acting on primitive stem cells.

Previously, we serially transplanted tumors that produced colony-stimulating factor (CSF) into nude mice, who developed marked granulocytosis along with tumor growth; their leukocyte counts reaching approximately one million per cu mm. The numbers of CFU-GM, CFU-E, CFU-Meg, CFU-S and BFU-E were increased in nude mice bearing CSF-producing tumor. We here report that tumor-conditioned medium (TCM...

Failure to demonstrate pluripotential hemopoietic stem cells in mouse brains.

Hemopoietic stem cells as defined by the capacity to produce spleen colonies in lethally irradiated recipients were reported by P. F. Bartlett [(1982) Proc. Natl. Acad. Sci. USA 79, 2722-2725] to be present in high frequencies in mouse brain. He also reported similar numbers of colony-forming units, spleen (CFU-s), in the brains of Wf/Wf mice, the bone marrow of which lacks detectable spleen co...

Comparative in vitro effects of cyclophosphamide derivatives on murine bone marrow-derived stromal and hemopoietic progenitor cell classes.

We investigated the in vitro effects of ASTA-Z-7595, ASTA-Z-7557, ASTA-Z-7654, and 4-hydroperoxycyclophosphamide (4HC) on murine stromal fibroblastoid colony-forming units, committed hemopoietic progenitors (erythroid burst-forming units and granulocyte/macrophage colony-forming units), and pluripotent hemopoietic stem cells assayed by the spleen colony-forming unit (CFU-s) assay. In general, t...
